The Structure of Actin-rich Filaments of Muscles According to X-ray Diffraction* by Cecily

چکیده
In the accompanying paper 1 we have described the structure of paramyosin in molluscan muscles. A second type of fibrous muscle component is possibly of greater significance, since it has been found in all muscles thus far examined, in smooth and striated varieties from vertebrate and invertebrate animals (Schmitt, Bear, Hall, and Jakus, 1947). When first discovered by means of x-ray diffraction this second component was designated type II (Bear, 1945). Subsequent x-ray examination of actin films by Astbury and Spark (1947; see also Astbury, 1947, 1949) indicated that it is an actin-rich fibrous system, if not composed of actin alone. In what follows for brevity we shall call this fibrous system actin, although very complete knowledge of the diffraction properties of isolated actin has not been available during most of this investigation, which deals only with results obtained from intact dried muscles. In the original diffraction work unambiguous index assignments could not be made for the observed reflections. I t was clear, however, that the axial identity period was in the range 350 to 420 A. Subsequent electron-optical investigations of striated muscle have revealed a similar axial periodicity (ca. 400 A, cf. Draper and Hodge, 1949), which has come to be considered, though without detailed justification, as related to the same system observed by the early diffraction studies. Recently Philpott and Szent-Gy/Jrgyi (1954)

The Structure of Actin-rich Filaments of Muscles According to X-ray Diffraction
From analysis of moderate- to small-angle x-ray diffraction patterns, in the light of similar experience with paramyosin, has been derived the following description for the structure of actin-rich filaments in "tinted" portions of the adductor muscle of the clam, Venus mercenaria: 1. Some 11 diffraction maxima, widely streaked along layer lines and occurring at moderate diffraction angles (spac...
متن کاملRecent X-ray Diffraction and Electron Microscope Studies of Striated Muscle
The sliding filament model for muscular contraction supposes that an appropriately directed force is developed between the actin and myosin filaments by some process in which the cross-bridges are involved. The cross-bridges between the filaments are believed to represent the parts of the myosin molecules which possess the active sites for ATPase activity and actin-binding ability, and project ...
متن کاملSynthesis of BiVO4 nanoparticles by the co-precipitation method and study the crystal structure, optical and photocatalytic properties of them
In this paper, the bismuth vanadate (BiVO4) nanoparticles were synthesized at 600 °C calcination temperature by co-precipitation method. To study the crystal structure, morphology, optical and photocatalytic properties of the samples, the X-ray diffraction analysis, field emission scanning electron microscopy, energy-dispersive X-ray spectroscopy, Fourier-transform infrared spectroscopy and ult...
متن کاملX-ray fiber diffraction modeling of structural changes of the thin filament upon activation of live vertebrate skeletal muscles
In order to clarify the structural changes of the thin filaments related to the regulation mechanism in skeletal muscle contraction, the intensities of thin filament-based reflections in the X-ray fiber diffraction patterns from live frog skeletal muscles at non-filament overlap length were investigated in the relaxed state and upon activation. Modeling the structural changes of the whole thin ...
متن کاملSurface Hardness Measurment and Microstructural Characterisation of Steel by X-Ray Diffraction Profile Analysis
An X-ray diffraction line will broaden considerably when steels change into martensitic structure on quenching. The results presented in this paper show that X-ray diffraction technique can be employed for a rapid and nondestructive measurement of hardness of hardened steel. Measurement on various quenched and tempered steels showed that the breadth of its diffraction peak increased with increa...
